你第一part 用了unless, 未確實.  但 聽係確實, "我會代替他".  有少少唔match.
Can = 可以, 你offer比你boss一個option.
Will = ...

因為我以為將來一定用Will...

但係正路黎講唔好淨係用i will replace him，除非已經firm左你係替佢

因為你淨用will...即係已經決定左，但係呢D野...正路都係老細決定，你最多都係提出你可以替佢

Replace is not an appropriate  word to use in this situation.

my colleague will be on holiday next week.....

I can stand in for him.
I can substitute (pronounced subs  tit  tute) for him.
I can be his substitution. (formal)
I can cover for him ( casual )
